We’re using Exchange in front of Poste. Poste just received all unknown mailboxes relayed from exchange.

If i want to send a mail back from poste to same mail domain used on exchange I receive SMTP 550 Error: no such user

Is there a possibility to relay unknown mailboxes as well?

With postfix it will work but with dovecot I don’t know

Yes it is desired behaviour. If poste has defined that domain in database it will not look nor deliver elsewhere and will try to deliver localy only (smarthost will not be used). To do this kind of wizardry it would require routing customization in code.

It is possible to relay whole domains. Relay unkown mailboxes is not possible…
