Is it possible to configure the plugin in a way that the unapprove functionality is enabled by default for new Projects?

Yes, this is possible in the latest version of the plugin.

You can enable the auto-unapprove plugin globally, which will ensure it is enabled by default for all projects. Project admins are then able to override the global setting if they wish, and repo admins are able to override the project setting.
